Justification: Hamsters will be provided with running wheels that are part of a 
computerized system that records and analyzes wheel revolutions. The 
circadian wheel running rhythm will be used as a marker for the timing of the 
underlying circadian clock, as is commonly done in many studies of circadian 
rhythms. As a result we will need to place hamsters in light-dark cycles when we 
study the synchronization of the clock with light and in constant darkness or 
constant light when we study the timing of the clock unrestrained by light-dark 
cycles.

Justification; To test the effects of visual information on development of the visual 
system and recovery from damage, some hamsters will be maintained in a light- 
tight room from birth to various ages (30, 60, 90, 120, and 300 days of age). Any

Justification: All of our breeding female Siberian dwarf hamsters with litters are 
housed with the adult male sire because our experience is that, unlike common 
golden hamsters, the Siberian dwarf hamster male is not injured by the female 
and he participates in the rearing of the young such that pup survival is higher 
when the sire is present. 

Number of animals affected: Approximately 2386 Siberian hamsters as it applies 
to this reporting period. This includes brewing pairs and their progeny. 

3.31fatf1) “Primary enclosures shall be cleaned and sanitized often enough to 
prevent an accumulation of excreta or debris: Provided, however, that such 
enclosures shall be sanitized at lease once every 2 weeks...” 

We sometimes do not change hamster cages for up to maximally 3 weeks due to 
the experimental needs of lACUC-approved research involving territorial 
aggression and circadian rhythms. 

Protocol A05017-0608 

JUSTIFICATION: The resident-intruder paradigm is a test used to study 
aggression in hamsters. When an unfamiliar hamster (“intruder”) is placed into 
the home cage of a “resident” hamster, the resident exhibits a great deal of 
aggression towards the intruder. Our lab injects drugs into various areas of the 
brain to study how neuropeptides influence these aggressive behaviors, in order 
to help residents establish their “territories” it is necessary that the bedding not 
be changed in the home cage of the resident for 1 week prior to testing. This 
leaves only 1 week for testing before the cages must be changed. However, we 
often require several test days (up to 4) in order to test several drugs or multiple 
doses of a single drug in the same animal. We usually wait 4-5 days between 
tests because many of these drugs remain In the animals’ system for several 
days. Thus, an additional week of “no cage changing” would allow sufficient time 
to conduct multiple tests of aggression under a variety of drug conditions. 
Importantly, our experiments require that this 3 week interval between cage

changes be imposed only once during the life of the hamster - during the 
aforementioned series of behavioral tests. 

Protocols A0601 2-1 208 &A06014-1 208 (both have identical justification as 
detailed below) 

JUSTIFICATION; I would like to extend the maximum time between cage 
changes to 3 weeks. This is not for routine cage changes, but for special 
experimental cases for which we are concerned that the odor stimulus provided 
by the odor-donor's cage is not sufficiently strong. This is the case when 
experimental animals are not allowed to contact the bedding stimulus (as part of 
an experimental manipulation) and thus do not activate additional nasal 
chemoreceptors. A maximum of 3 weeks between cage changes would allow for 
maximal odor accumulation in a stimulus animal's cage without causing 
substantial ammonia accumulation. 

In the past, we have acquired all of the animals in our colony from Charles River 
Laboratories with the exception of a small number of litters that we have bred 
ourselves. However, as a result of the success we have had in breeding and in 
an effort to control the developmental history of our animal subjects, we wish to 
breed and maintain a stable colony of Syrian hamsters at Georgia State 
University in lieu of ordering from an outside vendor. Care will be taken to 
reduce maternal stress both during pregnancy and after parturition, as this may 
cause her to spontaneously abort or cannibalize her offspring. We are 
requesting 3 weeks between cage changes for the breeding female. That is, the 
cage is changed 1 week prior to birth and then changed again 2 weeks after birth 
as we are currently approved for 3 week cage changes. 

Number of animals affected: Approximately 2463 as it applies to ail three 
protocols during this reporting period.

hamsters shall be at least 6 inches.” 

Protocol A07001-0210 

Justification: We maintain our Siberian dwarf in mouse cages, some of which are 
6 inches in height and some of which are 5 inches in height. All Turkish and 
Syrian hamsters are housed in cages which are 8 inches in height. We have 
found the mouse cages 5 inches in height to be of suitable height for the Siberian 
dwarf hamsters and thus continue this practice. 

Number of animals affected: Approximately 3605 are maintained in cages that 
are 5 inches in height as it applies to this reporting period. This represents 
approximately 75% of our Siberian hamster population. 

3.30 “Unless food supplements consumed by guinea pigs or hamsters supply 
them with their normal water requirements, potable water shall be provided daily 
except as might otherwise be required to provide adequate veterinary care.” 

Protocol A06014-1 208 

Justification: Electrophysiological recordings are performed in awake, behaving 
male hamsters that have been trained to poke their nose into a small port that 
provides them with exposure to different odorants. Upon detection of an odorant, 
the animal must hold its nose in the odor port for 1-1 .5 sec to receive water 
reward at a separate water dispenser. To maintain performance, hamsters are 
deprived of water for 24 hours prior to testing (as well as food, as rodents do not 
consume food without water). Animals will be allowed at least 48 hours of ad 
libitum access to food and water between deprivation periods. As animals will be 
given water reward during testing, food will also be available in the testing 
environment. Animals are monitored daily for dehydration by testing skin 
elasticity (pulled skin that fails to snap back to animal indicates dehydration) and 
are extensively handled prior to and after electrode implantation as well as 
between recording sessions in order to minimize stress to the animal. 

Number of animals affected: Approximately 240 as it applies to this reporting 
period.

3.29 “Guinea pigs and hamsters shall be fed each day except as othenwise 
might be required to provide adequate veterinary care.” 

Protocol A0501 7-0608 

JUSTIFICATION: The resident-intruder paradigm is a test used to study 
aggression in hamsters. When an unfamiliar hamster (“intruder”) is placed into 
the home cage of a “resident” hamster, the resident exhibits a great deal of 
aggression towards the intruder. However, approximately 25% of the residents 
that arrive from the vendor are not aggressive even after housing isolation. 

These animals are either removed from the study or become intruders for future 
studies, in order to standardize aggression and decrease the number of non- 
aggressive residents removed from the experiments, we would like to modify our 
resident intruder paradigm and improve our techniques to increase successful 
aggressive encounters. To do this, we would like to food deprive, food restrict, 
and/or alter dietary composition in addition to performing our normal resident- 
intruder protocol. If our preliminary studies demonstrate increased aggression 
with the food manipulations, we would use the protocol that would be least 
stressful for the animals. The protocol would be implemented for all the hamsters 
to keep the manipulations consistent. We hope to further increase aggressive 
behaviors through the use of food competition. Additionally, we would like to use 
these food intake paradigms to study the effect of competitive types of 
aggression on behavior and brain neurochemical changes in the animals 
subjected to the food manipulations. 

Food Deprivation: Resident and/or intruder hamsters may be subjected to food 
deprivation periods for up to 48 continuous hours prior to the resident-intruder 
paradigm. Water will be available ad libitum. It has been demonstrated that a 48- 
hour food deprivation does not produce detrimental, long-term changes to 
behavioral or physiological measures in hamsters [1]. Body weights and overall 
well-being will be monitored daily throughout the food deprivation periods. 
Records of the daily body weight measurements and changes will be kept by the 
researcher. Body weights will be recorded daily by the researchers and 
documented in the lab notebooks. A sheet will also be attached to the animal’s 
medical record clipboard listing the current body weights of the day. If any animal 
should lose more than 20% of their initial body weight, ad lib access to food and 
water will be provided until body weight loss is less than 10% of the initial start 
weight before being returned to the study. Under food deprivation, it takes 72

hours for Syrian hamsters to lose ~20% of their initial body weight [2]. The 48- 
hour food deprivation timeline is well below or within the acceptable range of 
what is normally perfonned for hamsters [1-3]. 

Food Restriction: Resident and/or intruder hamsters may be subjected to food 
restriction periods for up to 48 continuous hours prior to the resident-intruder 
paradigm. Hanging steel food hoppers will be used to hold food to facilitate food 
intake measurements. Animals will be given half the weight (approximately 5 
grams) of their normal ad libitum food intake. Body weights and overall well-being 
will be monitored throughout the food deprivation period. If any animal should 
lose more than 20% of their initial body weight, ad lib access to food and water 
will be provided until body weight loss is less than 10% of the initial start weight 
before being returned to the study. Under food-restriction, it takes 4-7 days 
before a 20% loss in body weight occurs, thus the 48 hours of food restriction are 
well within acceptable ranges [2, 3). 

Altered Dietary Composition: Resident and/or intruder hamsters may be 
subjected to alterations in their daily diets to promote food competition. Standard 
laboratory chow as supplied by animal facilities and/or nutritionally complete, 
well-balanced diets which have varying fat-carbohydrate contents will be 
provided for the hamsters prior to and during the resident-intruder pairing. 
Palatable foods and liquids (e.g. sucrose pellets [4], graham cracker crumbs[5], 
sweetened milk formula [3, 6]) will potentially be provided to the hamsters with 
the goal to study food competition without food deprivation. The intent is to 
provide these palatable items to the animals prior to and during the resident- 
intruder pairing. This is the alternative to food deprivation and food restriction. 
Additionally, the intention is to derive more data from the current experimental 
groups. However, this last food manipulation has the most potential to becoming 
an additional experimental objective. If so, this section will be amended further 
with more detail and animal numbers.

endogenous rhythms that are not entrained by the environmental lighting cycle. In order 
to assess the effects of drugs on endogenous circadian phase, the hamsters must be 
allowed to free-run under constant lighting conditions, in this case, constant darkness.”
